<Title>Innovative &#8220;factory-in-a-shoebox&#8221; could save lives</Title>

    <strong>Protein-building "factory-in-a-shoebox" could save lives worldwide</strong><br><br>By Sarah Hansen<br><br>The Technology Research Center (TRC) is a hidden gem at UMBC.  Tucked away just outside Hilltop Circle in a converted juvenile detention center, the place is brimming with interdisciplinary research projects poised to make powerful impacts on global health care.  Adil Zuber, a PhD student in the Frey Lab in UMBC’s Chemical, Biochemical, and Environmental Engineering Department, is immersed in one such project.  He’s working on a device that could revolutionize access to protein-based medications in remote or war-torn locales.   <br><br>“Imagine if you could make a device that could synthesize, and purify, and validate therapeutic protein on-site and on-demand within 24-48 hours.  And that’s what I’m working on,” Zuber says with pride.  <br><br>Therapeutic proteins are molecules that individuals need on a regular basis to maintain their health, and in some cases, to stay alive.  For example, diabetics need insulin.  Zuber’s project currently focuses on two others: erythropoietin (EPO) and streptokinase, although insulin is on the list for phase two of the project.  Streptokinase is used during heart attacks to dissolve blood clots and is on the World Health Organization’s “List of Essential Medicines.”  EPO regulates blood cell production and is used to treat anemia, a lack of sufficient red blood cells.  Sickle cell anemia is one of the most severe forms and can result in a wide array of symptoms including delayed growth and development.   The gene for sickle cell disease is most prevalent in Africa and the Middle East.<br><br>The device will be a godsend for people who need therapeutic proteins and find themselves in the midst of wars or natural disasters, where traditional supply lines are commonly disrupted and local stockpiles can be depleted quickly.  The factory-in-a-shoebox could also be used to produce “orphan drugs,” pharmaceuticals that no company wants to generate en masse because the demand is too small to make large-scale production profitable. <br><br>Zuber’s project uses cellular machinery to build these proteins, but without needing to support living cells.  Zuber’s collaborators at the biotech company Thermo Scientific are testing a process to break open cells and freeze-dry their contents for shipment with the devices.  Amazingly, upon reconstitution, the cell parts still function properly and happily build proteins within a mini-bioreactor.  Some bioreactors take up entire rooms, Zuber said, but these bioreactors are about half the size of a soda can and may be even smaller in the final product.  The whole process is called “Cell-Free Protein Synthesis.” Once the proteins are made, they need to be separated, or purified, from the rest of the molecular melee before they can safely be injected into a person.    <br><br>There are two steps to the purification process.  “Capture” separates the proteins from the bulk of their surrounding material, but isn’t precise enough to remove molecules too similar to the target protein, like target protein clumps or defective proteins.  Small segments of DNA can slip through the capture step, too.  So a second, finer-scale process called “polishing” is necessary.<br><br>Zuber has worked on both purification steps using different types of chromatography, a technique that separates molecules using thin columns about the size of a large pen.  The walls of the column are lined with molecules that bind to a tag on the target protein.  So, as the contents of the bioreactor flows through the column, the target protein stays stuck inside and the rest flows out the bottom of the column.  Then, the scientist changes the conditions in the column, such as its temperature or pH, so the target proteins will “let go” of the column wall and exit the tube for collection.<br><br>Zuber is testing a specific type of chromatography called “chromatofocusing” for use in the device.  Depending on the surrounding pH, a protein can be either positively or negatively charged.  At one specific pH, though, it will be neutral.  That pH is the protein’s “isoelectric point.”  At that point, the protein will detach from the column wall.  Chromatofocusing takes advantage of the isoelectric point to separate similar molecules, like a target protein and its misfolded counterpart, by changing the pH in a column in a carefully controlled way.  Zuber can separate molecules whose isoelectric points differ by as little as .1 pH.  Chromatography and chromatofocusing are established methods of purification, but the factory-in-a-box project subjects them to new constraints.            <br><br>While the project is technically challenging, “Part of what I love about it is that it’s so interdisciplinary and you’re really building soft skills,” said Zuber.  “Being able to communicate effectively with people across different disciplines, universities, and industries is critical.”  Zuber is collaborating with scientists, engineers, and regulatory experts at UMBC, the Ohio State University, and Thermo Scientific.  Each is working on a different aspect of the end product.<br><br>Collaboration is a boon for this complex project, but it necessitates frequent communication to help the team adhere to its aggressive timeline for completion.  “Everyone’s dependent upon each other,” Zuber said.  Because the goal is to create a marketable product, the team also has to meet high standards for manufacturing and production. <br><br>Zuber is excited by a vision of people using the factory-in-a-shoebox to provide medicines to children, the elderly, and others in dangerous and remote places around the world.  He knows there are plenty of other applications, too.  “I want to do something relevant, I want to do something important, I want to change the world,” he says.  “I feel I have the ability to make that impact, and that’s one of the most intellectually satisfying things about it.”<br>      <br>          <br><br>

<Title>Alternate Delivery Program Call for Participation Released</Title>
<Tagline>Hybrid/Online Course Redesign for Summer and Winter Courses</Tagline>

    <div class="html-content">Faculty who are interested in effective hybrid course delivery are invited to submit a proposal
        for this innovative faculty support program.<br>
        <br>
        The <a href="http://doit.umbc.edu/itnm/adp/" rel="nofollow external" class="bo">Alternate Delivery Program (<span>ADP</span>)</a> offers technical, pedagogical, and financial support to a selected cohort of faculty members who agree to convert an existing in-person course to hybrid delivery mode and offer that course in Summer Session 2015.  The 2015 <span>ADP</span>
        will begin in January, 2015 with a two day workshop for faculty (<a href="http://my.umbc.edu/groups/doit/events/27269" rel="nofollow external" class="bo">January 14th</a> and <a href="http://my.umbc.edu/groups/doit/events/27270" rel="nofollow external" class="bo">January 15th</a>) and the course conversion process will continue throughout the spring
        semester with technical support from DoIT's <a href="http://my.umbc.edu/groups/doit/news/45874" rel="nofollow external" class="bo">Instructional Technology Team</a>. 
        Proposals will also be considered from faculty who wish to convert a
        course to hybrid delivery and offer that course in Winter Session

<Title>Researcher of the Week: Alex Eftimiades</Title>

    <div>He is a Physics and Mathematics major. He is also a URA Scholar. His future plans includes obtaining his PhD in physics and focusing on research in computational physics.</div>

    <div><strong>How did you find your mentor for your research?</strong></div>
    <div>I told my adviser my interests and asked him who in the physics department shared those interests. He told me my best bet was the professor that I am now working with.</div>
    <div><br></div>
    <div><strong>How did you know this was the project you wanted to do?</strong></div>
    <div>I explained my interests and skill set to the professor I am now working with, and we discussed a possible research topic and the ways I could contribute. This particular project was a good fit for me because it satisfies my interests in the mathematics of curved space-time and quantum field theory, and it makes use of my programming experience.</div>
    <div><br></div>
    <div><strong>Is this your first independent research project?</strong></div>
    <div>No. I often spend my free time exploring my interests in the form of independent projects.</div>
    <div><br></div>
    <div><strong>How did you hear about the Undergraduate Research Award (<a href="http://www.umbc.edu/undergrad_ed/research/URA/" rel="nofollow external" class="bo">URA</a>) program?</strong></div>
    <div>Since I spend so much time on independent projects, I had asked Janet McGlynn about ways I might get funding for one of them long before I applied for funding for the project I am working on today.</div>
    <div><br></div>
    <div><strong>What academic background did you have before you applied for the URA?</strong></div>
    <div>I had spent a lot of time independently studying the subject I am now researching before applying for the URA. This included reading-both directed and undirected--and writing my own simulations of phenomena similar to the one that I am now investigating.</div>
    <div><br></div>
    <div><strong>Was the application difficult to do?</strong></div>
    <div>Not particularly. There were plenty of opportunities for getting feedback from Janet McGlynn and the application itself was pretty straightforward anyway.</div>
    <div><br></div>
    <div><strong>How much did your mentor help you with the application?</strong></div>
    <div>He mainly reviewed it for factual accuracy.</div>
    <div><br></div>
    <div><strong>What has been the hardest part about your research?</strong></div>
    <div>There are only a few good introductions to the topic I am investigating, and each one seems to provide a different piece of the whole picture. The hardest part so far has been putting those pieces together.</div>
    <div><br></div>
    <div><strong>What is your advice to other students about getting involved in research?</strong></div>
    <div>I have talked with a lot of friends about applying for research, and their biggest concern is consistently not feeling qualified. I have found applying for research to be like asking someone out on a date. You should be confident, and you should do it because of a genuine interest, not just for something to put on your resume. A lot of people ask for work in order to explore their interests, but I have found it best to explore your interests first. I only look for research on topics that I have explored on my own enough to conclude that I will stay interested, and in which I have enough background knowledge to be able to discuss possible research topics with a professor.</div>
    <div><br></div>
    <div><strong>What are your career goals?</strong></div>
    <div>I am most interested in getting my PhD in physics and doing research in computational physics. There are several areas I could see myself specializing in. I am also open to going into industry after I graduate if I can find a job doing something in line with my interests.</div>

    <div><strong>Did you transfer to UMBC from another institution? Where?</strong></div>
    <div>I transferred from the University of Maryland at College Park.</div>

    <p><strong>Q: What do you love most about UMBC?</strong><br> I love working with amazing, smart, diverse group of people to create new programs and events. I have always been encouraged to think out of the box and try new things to advance the mission of the University.</p>
    <p><strong>Q: What’s your favorite thing about your job?</strong><br> I enjoy providing opportunities for people to financially give back to UMBC and to see the impact of their giving by meeting their scholars or seeing how their support is making a difference at UMBC.</p>
    <p><strong>Q: Why do you think it’s important to support UMBC?</strong><br> We are educating the next generation of leaders in science, technology, education, arts, social science, and math. UMBC is making a difference. It is a great investment.</p>

<Title>Meet the Staff: Kim Robinson</Title>

    <p><em>Here in the Office of Institutional Advancement, we are always working hard to support the university and connect our donors with causes they care most about. We would like to introduce our donors — to whom we are so grateful — to the people who help make those things happen. Today, we’ll be hearing from <strong>Kim Robinson</strong>, the Senior Director of Development.</em></p>
    <p><strong><a href="https://umbcgiving.files.wordpress.com/2014/04/kim.jpg" rel="nofollow external" class="bo"><img src="https://umbcgiving.files.wordpress.com/2014/04/kim.jpg?w=225&amp;h=300" alt="kim" width="225" height="300" style="max-width: 100%; height: auto;"></a>Name:</strong> Kim Robinson<br> <strong>Job Title:</strong> Senior Director of Development<br> <strong>Focus Area:</strong> I enjoy meeting with alumni, retired faculty &amp; staff<strong>,</strong> and friends to reconnect them back to UMBC.  It is very rewarding to show the all the exciting things that are happening at UMBC and providing them with opportunity for them to make a difference in students’ lives and faculty research. I also oversee the planned giving program at UMBC.<br> <strong>Years at UMBC:</strong> 24 years</p>
    <p><strong>Q: Where are you from originally?</strong><br> Kentucky.</p>
    <p><strong>Q: What do you love most about UMBC?</strong><br> I love working with amazing, smart, diverse group of people to create new programs and events. I have always been encouraged to think out of the box and try new things to advance the mission of the University.</p>
    <p><strong>Q: What’s your favorite thing about your job?</strong><br> I enjoy providing opportunities for people to financially give back to UMBC and to see the impact of their giving by meeting their scholars or seeing how their support is making a difference at UMBC.</p>
    <p><strong>Q: Why do you think it’s important to support UMBC?</strong><br> We are educating the next generation of leaders in science, technology, education, arts, social science, and math. UMBC is making a difference. It is a great investment.</p>
    <p><strong>Q: What is your favorite food?</strong><br> Lobster.</p>
    <p><strong>Q: What is your favorite hobby or pastime?</strong><br> I spend a lot of time at athletic facilities. I enjoy watching my children play sports (soccer, lacrosse, baseball, volleyball, and basketball) and spending quality time with family and friends. I love the beach and water activities.</p>
