Stories of a Big City Farm Girl, Vol. 2

Editor’s Note: This is the second in a series of three posts from Lexi, our summer intern here in the Alumni Communications office, about her experiences as a new-to-Baltimore student.

In my last post, I described how different the driving conditions in Baltimore are from rural Delaware. Multiple lane highways, aggressive drivers, lots of honking, and mis-informed GPSes always created interesting adventures.

But did you know that in Delaware, a lot of high schools don’t require you to learn how to parallel park during driver’s ed? Well, Milford High School didn’t at least. We just had to be able to park between the lines, navigate the bumps of the backroads, and not panic when passing the chicken trucks or tractors on the highway. The horrors of parallel parking didn’t hit me until college, as I had never driven in any type of city conditions, let alone tried to park sideways in between two cars that were worth more than my entire college education.

    photo via TripAdvisor

My first “lesson” came when I had to go to a museum for an art class. There was a definite trend with my art class excursions causing me more trouble than they were worth. I had to go to the Walters Art Museum in Mount Vernon. I knew I’d only be there for about an hour, and there was no way I was going to pay for the parking garages in Baltimore, so street parking was my only option. After circling the area and managing to go down every one-way street there was in
Mount Vernon, I found a spot. It was a
parallel spot, and on the left side. I was ready.

I stopped my car, put on my blinker, and then panicked. I had never parallel parked before, let alone parked on the left side of the street. Thank goodness there wasn’t any traffic. I gently tapped my accelerator to back up, but I had turned the wheel too far. I had turned my car in a little arch in the middle of the street. I pulled up next to the compact car in front of me, and tried again. This time, I managed to get almost all the way into the spot, but didn’t turn the wheel enough, and again sat almost in the middle of the street. Third time’s the charm, right? Mirror to mirror, I backed my car into the spot, one inch at a time. But this time, I had it. I could feel it. My car glided into the spot with no problems, I straightened it out, put it in park, and screamed.

I had never been more proud of myself in my life (OK, that’s a slight exaggeration, but I was very happy). I began congratulating myself, turned up the radio, sang along a little bit to Train’s “Hey Soul Sister,” and then realized my windows were down and people were staring. Blushing and laughing, I calmly got out of the car and went to pay for my spot. I was still so excited and embarrassed that I couldn’t get money out of my wallet. But, I thought, you just parallel parked all by yourself.

When I got back to my dorm, I told one of my friends, who then laughed at me, and made me go back outside and learn how to parallel park. While he sat next to me, his roommate sat in the back, and they both took turns shouting directions at me as I tried to recreate my parking skills in the back road next to Potomac Hall. Cars drove by, we all kept yelling, people stared, and we all got frustrated. But, in the end, I had learned how to parallel park. By the time we were finished, I couldn’t help but think, When I go home for the summer, I’m going to forget how to do all of this.

Summer research program fuels undergrad’s fire for biology

by Sarah Hansen

As an Amgen Scholar, UMBC senior Alana Lescure participated in an intensive nine-week research experience at the Massachusetts Institute of Technology this summer. Only 25 of 1000 applicants secured a position at MIT. For Lescure, the program was transformative. It opened her eyes to careers she hadn’t considered, connected her with elite researchers, and intensified her passion for biology. But there were less grandiose takeaways, too. She learned she needed to improve her pipetting skills.

Lescure worked in Rudolph Jaenisch’s lab under the guidance of a postdoc, Yun Li. Jaenisch is a professor and member of the prestigious Whitehead Institute at MIT, and his lab is “pretty intense,” said Lescure. “Everybody is very focused on their work.” Some of them are essentially nocturnal, but Lescure managed to maintain a normal schedule. She played a role in several projects in the Jaenisch lab. For one, she helped analyze cerebral organoids, groups of nerve cells induced to grow into three-dimensional structures nicknamed “mini-brains.” She looked at images taken by a high-resolution microscope to determine what kinds of cells were developing in the organoids. “You get these beautiful images. Even if they can’t tell you anything, they’re so beautiful,” she said.

Her main accomplishment, though, was creating a plasmid, a small circular piece of DNA, which carries a specific gene with a mutation. The mutation is associated with autism spectrum disorders and a range of other problems such as seizures, intellectual disability, and delayed development.

Lescure started with a plasmid “backbone,” a DNA circle that did not contain the mutated gene yet. Her job was to use molecular techniques to add the gene of interest, called MECP2, to the backbone.

“It took me all summer to get [the plasmid], because the backbone I was working with was really stubborn,” she said. Once she finally created the plasmid, she inserted it into bacterial cells for cloning. Each time the bacteria divide (which can be every twenty minutes under ideal conditions), they copy all of their own DNA and the added plasmid. The goal of cloning is to create lots of copies of the mutated gene.

After she cloned the plasmid into bacterial cells and they divided many times, she had to get the plasmid DNA back out of the bacteria (it was only there to multiply). Once she finished the purification process, it was time for Lescure to come home. Li will transfer the mutated gene to human embryonic stem cells and then induce those stem cells to grow into different types of brain cells – also no easy task. They’re generating brain cells with the mutation because the brain produces a lot of MeCP2 protein.

“Hopefully in a couple months I can contact them again to see how it’s going,” Lescure said. But her summer at MIT was about a lot more than manipulating DNA. She regularly left the lab to attend seminars given by big names in science. As a Biological Sciences major at UMBC, she’s read plenty of scientific papers, but this summer, “I actually got to hear some of them speak about their research,” she said.

Lescure brought confidence to her work in Jaenisch’s lab, partly because of the molecular biology research experience she already had from Lasse Lindahl’s lab here at UMBC. But even she got a little nervous when surrounded by Nobel laureates and others with mile-long resumes. “I felt like I shouldn’t be allowed to interact with these people,” she said. She overcame her fear, though. On an all-expenses-paid weekend trip to University of California at Los Angeles for all the Amgen Scholars (there were 250 nation-wide), she snatched opportunities for one-on-one conversations with top-shelf scientists. Linda Liau, professor of neurosurgery at UCLA and neurosurgeon at UCLA Medical Center, has been featured on television for her work. “It was so inspiring to see somebody who started out as an undergrad researcher and now has this great career,” Lescure said.

Cen Xu, Scientific Director at Amgen, also made a big impact. Talking to Xu taught Lescure about different applications for basic research that uncovers potential drug targets. “That really inspired me and got me interested in neuroscience.”

The Amgen program also included panel discussions that taught Lescure there are career paths other than academia or industry that would take advantage of her passion for science. “I started to get ideas that there are so many options. That I don’t have to just go and be a researcher. That there are lots of other possibilities,” she said.

She’s still shooting for graduate school after she graduates from UMBC in December, though. The Amgen Scholar experience only pushed her farther in that direction. “It was a really good experience. It definitely gave me more of an idea of where I want to go. It got me really excited about science.”
